Seeing that the "tech specs" section of the QuickBooks Enterprise 9.0 web site didn't mention anything about compatibility with 64 bit computers, I did some digging directly with Intuit.
A thank you to Rich Walker from Intuit for coordinating directly with the Enterprise support team and for getting feedback for me on this issue.
In essence here is what you need to know - QuickBooks Enterprise 9.0 will run on 64 bit desktop and server computers. In fact, Intuit has many successful installations in these environments. According to Rich, full support is provided for these 64 bit installations of QuickBooks Enterprise 9.0. The Enterprise support team will address questions and help resolve customer issues in these environments, just as they do in the standard 32 bit computing environments.
I have asked Rich to provide some insight on Intuit's position relating to the use of QuickBooks Enterprise in a virtual server environment, and I hope to have additional insight on that issue soon.

Based on the information I have from Intuit, only Enterprise 2009 is supported in the 64 bit world. With the release of QuickBooks 2010 just around the corner, I don't yet know if that support will be extended to the Premier product line or not.
I have 2 clients that are currently in the SBS 2008 + 64 bit world running QuickBooks Enterprise with great success. They had some other issues with the server and 64 bit, but those issues were not related to the operation of QuickBooks.
Remember, I can offer you significant savings on Enterprise should you opt to go that route for your new server.

Can you provide any guidance for users of QB Premier 2009, vs. Enterprise? We're considering migrating our SBS 2003 x32 server--running the QB database--to SBS 2008 x64. It would be nice to know whether QB will work on the new server....

We have Quickbooks Enterprise running on a Dell PowerEdge 2900, 8 gigs of ram, 1 quad core processor (with a socket for another) and 3, 147 gig drives with raid 5. We are using Microsoft Server 2008 64-bit with Terminal Services. We LOVE the combination. Fast and reliable.

When you install Enterprise 9 on a 64 bit Vista system you must be sure to update to the latest revision. The earlier revisions, which you usually get when you do the install, will have problems with anything related to the PDF driver. This includes reconciling accounts and sending invoices (etc.) via email. The latest revision fixes these problems.
